本科生课程《蛋白质化学与工程》,研究生课程《蛋白质结构与现代药物开发》。指导学生获得福建省研究生优秀学位论文。

科研简介:

要从事蛋白的结构和功能研究,并基于结构进行多肽和蛋白类药物的改造和评价。目前已发表SCI论文九十余篇,先后主持国家自然科学基金面上项目及福建省自然科学基金重点项目等。主要研究领域包括:

1、肿瘤迁移相关蛋白的结构和功能研究:几乎90%癌症病人的死亡是由于癌细胞的转移性扩散引起。目前针对原发性肿瘤的治疗卓有成效,但靶向肿瘤转移方面的进展却不尽人意。我们主要针对肿瘤迁移相关的一些蛋白如尿激酶受体系统和多种丝氨酸蛋白酶,及其与抑制剂的复合物进行X-射线晶体结构研究,并针对这些结构指导抑制剂和靶向弹头的改造,以发展高效特异的肿瘤治疗药物。

2、血栓形成相关蛋白的结构和功能研究:人体保持血液处于正常流动状态,主要依赖于凝血系统和纤溶系统处于一个动态平衡。病理条件下两个系统的功能异常将导致出血或血栓形成。我们针对凝血和纤溶相关的一些蛋白如纤溶酶原激活物tPA、多种凝血因子及它们的调控蛋白PDI进行结构和功能的研究,并基于这些结构进行蛋白质工程蛋白及抑制剂的设计和改造,以发展高效且无出血副作用的溶栓和抗血栓形成药物。

3、光动力抗肿瘤和抗菌研究:光敏剂在特定波长的光激发下可产生具有生物毒性的单线态氧或者自由基,进而杀死细胞或微生物,可用于抗肿瘤和抗菌方面的治疗。传统的光敏剂存在靶向性差,水溶性不佳等问题。我们对光敏剂进行一系列的修饰和包裹,发展高靶向性光敏剂,用于抗肿瘤和耐药菌的治疗。
